摘要:硝酸盐在植物体内积累过多已成为影响蔬菜品质并影响人类健康的重要因素,硝酸还原酶是硝酸盐代谢中的关键酶,提高其活性有利于硝酸盐的降解.本文采用离体法测定不同条件对于茎瘤芥硝酸还原酶活性的影响.茎瘤芥硝酸还原酶经过不同的pH、不同的温度、不同的时间、不同的硝酸钾浓度以及硝酸还原酶不同的浓度的处理表明:茎瘤芥硝酸还原酶的最适pH为6.8;最适温度为25℃;最适底物硝酸钾浓度为mol/L,在茎瘤芥硝酸还原酶相对浓度足够大时,随着底物浓度增加,茎瘤芥硝酸还原酶的活性逐渐上升.反应条件超过或者达不到最适条件时酶活性都会受到抑制.同时,在底物硝酸钾浓度足够大时,酶液相对浓度较低的情况下,逐渐增加酶液相对浓度,反应速度随着硝酸还原酶相对浓度的增加而增加,酶反应速率也随之增加.
